Introduction of Mutagenesis Technology

Mutagenesis induces mutations in the genes of strains by artificial measures, mainly chemical mutagenesis, physical mutagenesis and site-directed mutagenesis. Induced mutations produced by physical or chemical mutagens are changes in genes after contact with mutagens and environmental causes that produce non-site-specific random mutations. Conventional mutagenesis methods are to some extent easy to perform and can increase the rate of resistance mutations. For example, Lactobacillus casei LC2W was used as the starting strain for nitrosoguanidine and UV mutagenesis, and the mutagenic strains LC2W-NTG-12 and LC2W-UV-11 were screened to show high acid production and acid tolerance performance. Site-directed mutagenesis is a technique for creating a library of genetic information by introducing a mutation into a DNA sequence. This technique promotes improved strain performance by generating specific DNA mutations (including point mutations, deletion mutations and insertion mutations).

Our Methods

  • Chemical mutagenesis

Chemical mutagenesis mainly uses some chemical mutagens. Most chemical mutagens are base analogs, base modifiers and shift mutagens, including bromodeoxyuridine (BrdU), alkylating agents, agents that form DNA adducts, DNA intercalating agents, DNA crosslinkers, etc.

  • Physical mutagenesis

Physical mutagenesis methods mainly utilize UV, α-rays, β-rays and γ-rays. In addition, we also offer new physical mutagenesis methods based on atmospheric room temperature plasma (ARTP).

  • Site-directed mutagenesis

Site-directed mutagenesis is a method of mutating the DNA sequence of a strain. In the Creative Enzymes' lab, our scientists use molecular biology methods to synthesize short DNA primers that contain the desired mutation site and are complementary to the template DNA surrounding the mutation site. The single-stranded primers are then extended using DNA polymerase, and the replicated gene contains the mutation site. Mutations can be single base changes, multiple base changes, deletions or insertions.
